Seed Tissue Structure

Seed tissue structure is the organized arrangement of protective, nutritive, and embryonic tissues that enables a seed to survive dormancy and support a new plant. The seed coat shields internal tissues from physical damage and water loss, while the endosperm or cotyledons store carbohydrates, proteins, and lipids that nourish the embryo during germination; the embryo then resumes growth when moisture, temperature, and oxygen are suitable. Studying these tissues helps explain seed development, dormancy, germination, and seedling establishment. In biology and agriculture, structural analysis supports crop improvement, seed quality assessment, and research on plant reproduction and stress tolerance.

We present the procedure to assemble a seed coat bedding assay (SCBA) from Arabidopsis thaliana seeds. The SCBA was shown to be a powerful tool to explore genetically and in vitro how the endosperm controls seed germination in dormant seeds and in response to light cues. The SCBA is in principle applicable under any situation where the endosperm is suspected to influence embryonic growth.
