Serotype Performance

Serotype performance is the comparative assessment of how distinct serotypes, or antigenically different variants of a microorganism or biological vector, behave under defined experimental conditions. Differences in surface molecules influence receptor binding, cell entry, replication or delivery efficiency, and recognition by antibodies, so researchers measure each serotype using standardized biological assays. In biology, these comparisons help identify serotypes best suited to infection studies, vaccine design, diagnostics, or gene-delivery systems. Evaluating performance alongside tissue specificity, immune response, and experimental reproducibility supports better understanding of host-pathogen interactions and guides the selection of effective, safer tools for research and therapeutic development.

Here, we present a protocol to generate a proof-of-principle divalent adenovirus type 5 (Ad5) vector Ad5/H5-HVR1-KWAS-HVR5-His6 by utilizing the Antigen Capsid-Incorporation strategy. This vector was demonstrated to exhibit qualitative fitness, the capability to escape Ad5-positive sera in vitro, and the antigenicity as well as immunogenicity to the incorporated antigens.
