Serum-free Incubation

Serum-free incubation is a cell or tissue culture method performed without animal serum, enabling more controlled biological experiments and reducing variability from undefined serum components. During incubation, cells remain in a basal medium supplemented with defined nutrients, hormones, growth factors, or other specified additives that support survival and regulate proliferation, differentiation, or signaling. This approach helps researchers isolate the effects of individual factors, improve reproducibility, and reduce interference from proteins and growth-promoting molecules found in serum. In biology, serum-free incubation supports cell-based assays, recombinant protein production, stem cell research, and studies of cellular responses under precisely controlled conditions.

Isolation of Small Noncoding RNAs from Human Serum

This protocol describes a method for extracting small RNAs from human serum. We have used this method to isolate microRNAs from cancer serum for use in DNA arrays and also singleplex quantitative PCR. The protocol utilizes phenol and guanidinium thiocyanate reagents with modifications to yield high quality RNA.

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.
