Siderophore Secretion

Siderophore secretion is a microbial process in which bacteria, fungi, and other microorganisms release specialized molecules that acquire iron from their surroundings, an essential function because iron is vital yet often poorly soluble and biologically limited. These compounds chelate ferric iron with high affinity, forming iron–siderophore complexes that cells recognize and transport across their membranes; intracellular processes then release the iron for metabolic use. In biology, siderophore secretion influences microbial growth, competition, host colonization, and interactions within communities. Studying this process supports research on nutrient acquisition, microbial ecology, pathogenicity, and potential strategies for antimicrobial development or beneficial plant–microbe interactions.

Source: Gupta, S., Marcela Rodriguez, G. Isolation and Characterization of Extracellular Vesicles Produced by Iron-limited Mycobacteria. J. Vis. Exp. (2019)This video demonstrates the stepwise isolation of extracellular vesicles (EVs) from Mycobacterium tuberculosis cultured under iron-limiting conditions that mimic the host environment. It outlines the use of ultrafiltration, differential centrifugation, and density gradient separation to enrich and identify EV-containing fractions.

This protocol describes how to perform rapid low-cost luciferase assays at medium-throughput using an insulin-linked Gaussia luciferase as a proxy for insulin secretion from beta cells. The assay can be performed with most luminescence plate readers and multichannel pipettes.

Functional Assessment of Antibody-Secreting B Cells Using ELISpot Assay

2025

This video demonstrates the use of the ELISpot assay to assess the function of antibody-secreting B cells, ASCs. The antibodies secreted by the ASCs interact with the membrane-bound polyclonal antibody fragments, which are detected by the enzyme-conjugated IgG or IgM antibodies. The colored spot produced in each well indicates the presence of the specific antibody's isotype, which correlates with the B cells' secretory activity.

Source: Tonya J. Webb1 1 Department of Microbiology and Immunology, University of Maryland School of Medicine and the Marlene and Stewart Greenebaum Comprehensive Cancer Center, Baltimore, Maryland 21201 ELISPOT is a standardized, reproducible assay used to detect cellular immune responses. The assay utilizes an enzyme-linked immunosorbent assay (ELISA)- based method to detect single-cell immune responses which can be visualized by spots, hence the name ELISPOT. ELISPOT was first described in...
