Sinoatrial Node

The sinoatrial node is a specialized group of cardiac cells in the right atrium that normally initiates each heartbeat, making it the heart’s natural pacemaker. Its pacemaker cells spontaneously depolarize because of gradual ion movements across the cell membrane, including the hyperpolarization-activated “funny” current and calcium influx, until they generate an action potential. This electrical signal spreads through the atria and then reaches the atrioventricular node, coordinating the sequence of cardiac contraction. Studying the sinoatrial node helps explain heart-rate regulation by the autonomic nervous system and supports research into arrhythmias, conduction disorders, and artificial pacemaker design.

This video demonstrates the procedure to isolate sinoatrial node or SAN from mouse heart. The SAN houses the sinoatrial cells that initiates heartbeat by generating spontaneous action potentials.

Methods for the Isolation, Culture, and Functional Characterization of Sinoatrial Node Myocytes from Adult Mice

Methods are demonstrated for the isolation of sinoatrial node myocytes (SAMs) from adult mice for patch clamp electrophysiology or imaging studies. Isolated cells can be used directly or can be maintained in culture to permit expression of proteins of interest, such as genetically encoded reporters.
