Small-scale Preparation

Small-scale preparation is the controlled processing of limited quantities of biological samples, cells, or reagents for laboratory analysis and experimentation. It typically uses microscale volumes, precise pipetting, and selective steps such as homogenization, centrifugation, filtration, or extraction to isolate and concentrate the material of interest while minimizing sample loss and reagent use. In biology, these preparations support tasks ranging from nucleic acid and protein analysis to enzyme assays and preliminary method development. Working at small scale can reduce costs, conserve scarce samples, increase experimental throughput, and help researchers optimize protocols before applying them to larger or more complex biological systems.

Reliable, intermediate scale preparation of 1,2,3,4,5-pentamethylcyclopentadiene (Cp*H) is presented. The revised protocol for the synthesis and purification of the ligand minimizes the need for specialized laboratory equipment while simplifying reaction workups and product purification. Use of Cp*H in the synthesis of [Cp*MCl2]2 complexes (M = Ru, Ir) is also described.

This article presents a small-scale plasma membrane isolation protocol for the characterization of Candida albicans ABC (ATP-binding cassette) protein Cdr1, overexpressed in Saccharomyces cerevisiae.
