Brown Adipose Tissue

Brown adipose tissue is a specialized fat depot that converts stored chemical energy into heat, helping maintain body temperature and regulate whole-body energy balance. In response to cold or sympathetic nervous system signals, norepinephrine stimulates brown adipocytes, where mitochondria use uncoupling protein 1 to dissipate the proton gradient generated by the respiratory chain as heat rather than producing ATP. This non-shivering thermogenesis is especially important in newborns and small mammals, while adults retain smaller, inducible brown-fat depots. Studying brown adipose tissue may clarify energy metabolism and support research on obesity, diabetes, cold adaptation, and metabolic imaging.

Here we describe a protocol for efficient chromatin immunoprecipitation (ChIP) followed by high-throughput DNA sequencing (ChIP-seq) of brown adipose tissue (BAT) isolated from a mouse. This protocol is suitable for both mapping histone modifications and investigating genome-wide localization of non-histone proteins of interest in vivo.

The method presented here uses 18F-Fluorodeoxyglucose (18F-FDG) positron emission tomography/computed tomography (PET-CT) and fat-water separated magnetic resonance imaging (MRI), each scanned following 2 hr exposure to thermoneutral (24 °C) and cold conditions (17 °C) in order to map brown adipose tissue (BAT) in adult human subjects.
