Sputum Decontamination

Sputum decontamination is a laboratory preparation method that reduces unwanted microorganisms and mucus in respiratory specimens, improving the recovery and identification of clinically important pathogens. In a common approach, N-acetyl-L-cysteine breaks disulfide bonds in mucin to liquefy sputum, while sodium hydroxide suppresses many contaminating bacteria; centrifugation can then concentrate the remaining organisms for analysis. The procedure is especially important when culturing or detecting mycobacteria, whose slow growth makes them vulnerable to overgrowth by normal respiratory flora. Properly controlled decontamination supports more reliable microscopy, culture, and molecular testing while preserving viable target organisms.
