Mid-late Instar

Mid-late instar refers to the middle-to-advanced portion of an insect or other arthropod’s developmental stage between two successive molts, when the animal grows substantially before entering its next form. During this period, cells and tissues increase in size, nutritional reserves accumulate, and hormonal signals coordinate preparation for ecdysis, the shedding of the old cuticle; the new cuticle then expands and hardens. Identifying mid-late instars helps researchers stage specimens consistently, compare growth and development, and select appropriate ages for studies of physiology, behavior, genetics, pest control, and environmental responses. The term is especially useful when development proceeds through multiple larval or nymphal instars.

The Utility of Stage-specific Mid-to-late Drosophila Follicle Isolation

Stage-specific isolation of mid-to-late Drosophila follicles is useful for a variety of purposes. Such follicles develop in culture, which allows for genetic and/or pharmacologic manipulations to be coupled with in vitro development assays and live imaging. Additionally, follicles can be used for molecular studies, such as isolating mRNA and protein.

Insecticide evaluations are often targeted against adult insects, rather than immature stages. Here, we present a protocol for evaluating insecticides against bed bug eggs with a comparison to the first nymphal bed bug stage. These protocols could be adjusted for other insects to evaluate insecticide efficacy in non-adult life stages.

Measuring left ventricular pressure (LV) in embryonic and neonatal mice is described. Pressure is measured by inserting a needle connected to a fluid-filled transducer into the LV under ultrasound guidance. Care must be taken to maintain normal cardiac function during the experimental protocol.
