Stem Cell Factors

Stem cell factors are molecular signals and regulatory proteins that maintain stem cell identity, promote self-renewal, or direct differentiation, making them central to understanding how tissues develop and repair. Transcription factors such as OCT4, SOX2, and NANOG regulate gene expression networks, while extracellular cues including growth factors activate signaling pathways that alter cell state according to surrounding conditions. In biology research, these factors help researchers control embryonic, adult, and induced pluripotent stem cells for studying development, modeling disease, testing therapies, and engineering regenerative tissues. Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
