Stem Cell Zone Model

The Stem Cell Zone Model is a biological framework that explains how the location of stem cells within a tissue influences their maintenance, behavior, and fate. It proposes that spatially distinct zones provide different combinations of signaling molecules, neighboring cell interactions, and extracellular matrix conditions, creating local environments that regulate self-renewal or promote differentiation. In biology, this model helps connect stem cell niches with tissue organization and homeostasis. Researchers use it to interpret how tissues regenerate, how stem cell populations respond to injury, and how disrupted microenvironments may contribute to disease or altered developmental processes.

Establishing culture systems for the expansion of adult neural stem cells (aNSCs) allows for the examination and application of aNSCs for therapy. The subcallosal zone (SCZ) has recently been recognized as a novel neuroblast-forming region in adult mice. Here, methods for the isolation, expansion, and differentiation of SCZ-aNSCs are described.

Isolation and Culturing of Adult Neural Stem Cells from the Mouse Subcallosal Zone

The video demonstrates the process of isolating and culturing neural stem cells from the mouse brain's subcallosal zone. It involves dissecting tissue, enzymatic dissociation, and culturing them with differentiation factors to develop into mature neural cells.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

2015

We report a Fluorescent Activated Cell Sorting (FACS)-based method to isolate neural stem cells (NSCs) and their progeny from the subventricular zone (SVZ) of the adult mouse brain. Applied to Fluorescence Ubiquitination Cell Cycle Indicator (FUCCI) transgenic mice, it allows the study of cell cycle progression by live imaging.
