Substrate Uncoupler Inhibitor

Substrate, uncoupler, and inhibitor compounds are experimental tools used to dissect cellular energy metabolism, especially mitochondrial respiration. Substrates provide electrons that enter the electron transport chain, uncouplers dissipate the proton gradient across the inner mitochondrial membrane without directly stopping electron flow, and inhibitors block defined respiratory complexes or transport steps. By applying these compounds in controlled sequences, researchers can distinguish oxygen consumption driven by substrate oxidation from proton-leak respiration and determine how efficiently mitochondria generate ATP. This approach supports studies of bioenergetics, metabolic disease, toxicology, drug action, and cell-state changes in biology.

Modelling Intratumoral Treatment with miRNA Inhibitors: A Method to Analyze the Effect of miRNA Inhibitors on Tumor Growth in Murine Model

2023

This video demonstrates the in vivo delivery of miRNA inhibitors as therapeutic agents against tumor growth in orthotopic mouse models. The intratumoral administration of synthetic miRNA inhibitors or antagomiRs allows them to bind and inhibit endogenous miRNAs, thereby causing mRNA upregulation and suppression of tumor growth.

Eukaryotic Transcription Inhibitors

2020

Certain biochemical processes, such as embryonic development and cell growth regulation, depend on the repression of specific genes. DNA binding proteins known as eukaryotic transcription inhibitors regulate the repression of gene expression in eukaryotes. The presence of these inhibitors at the required location and time in the cell is triggered by the presence of hormones and additional signals from other cells. Eukaryotic transcription inhibitors usually contain two distinct domains, a DNA...

Source: Guo, S., et al. Quantifying Bacterial Surface Swarming Motility on Inducer Gradient Plates. J. Vis. Exp. (2022).This video demonstrates the use of a double-layer gradient swarm plate to study bacterial swarming behavior under varying inhibitor concentrations. A plate with a gradient-forming inhibitor in the bottom layer is inoculated with bacteria and incubated. Images are captured at regular intervals to analyze the effect of inhibitor concentration on bacterial swarming.
