Stopped Flow Spectrofluorimetry

Stopped-flow spectrofluorimetry is a rapid kinetic technique that measures changes in fluorescence immediately after two or more solutions mix, allowing researchers to observe fast molecular events in real time. In the instrument, reactants are driven into an observation cell, rapidly mixed, and then halted; excitation light stimulates fluorescent molecules or probes while emitted light is recorded as a time-dependent signal. Analysis of the resulting fluorescence traces reveals reaction rates, conformational changes, binding events, and intermediate states. In biology, the method helps characterize enzyme catalysis, protein folding, ligand interactions, and nucleic-acid processes that occur too quickly for conventional, manually sampled measurements.

Flow Cytometry Purification of Mouse Meiotic Cells

An efficient method to obtain highly purified viable meiotic fractions from mouse testis is described, which combines a refined cell dissociation protocol with fluorescent activated cell sorting (FACS). This method takes advantage of differences in the DNA content and nuclear density of discrete meiotic fractions.

Automation of the Micronucleus Assay Using Imaging Flow Cytometry and Artificial Intelligence

The micronucleus (MN) assay is a well-established test for quantifying DNA damage. However, scoring the assay using conventional techniques such as manual microscopy or feature-based image analysis is laborious and challenging. This paper describes the methodology to develop an artificial intelligence model to score the MN assay using imaging flow cytometry data.

The platelet adhesion cascade takes place in the presence of shear flow, a factor not accounted for in conventional (static) well-plate assays. This article reports on a platelet-aggregation assay utilizing a microfluidic well-plate format to emulate physiological shear flow conditions.
