Synovial Fibroblasts

Synovial fibroblasts are specialized connective-tissue cells that form much of the synovial lining surrounding diarthrodial joints, where they help maintain the extracellular matrix and support smooth movement. They produce joint-lining components such as hyaluronan and lubricating molecules, while responding to mechanical and inflammatory signals through changes in gene expression and cytokine secretion. When persistently activated, synovial fibroblasts can release inflammatory mediators and matrix-degrading enzymes that promote synovial hyperplasia, cartilage damage, and rheumatoid arthritis progression. Studying these cells helps explain joint homeostasis and chronic inflammation, supports disease modeling and biomarker research, and may identify targets for therapies that limit tissue destruction.

A 3D system of culturing human articular chondrocytes in high levels of synovial fluid is described. Synovial fluid reflects the most natural microenvironment for articular cartilage, and can be easily obtained and stored. This system thus can be used for studying cartilage regeneration and for screening therapeutics for treating arthritis.

Isolation and Cellular Phenotyping of Mesenchymal Stem Cells Derived from Synovial Fluid and Bone Marrow of Minipigs

A protocol establishing mesenchymal stem cells (MSCs) isolated from the synovial fluid and bone marrow of minipigs and non-invasively collected using syringe aspiration is presented. Cellular phenotyping was performed using flow cytometry after cell isolation and in vitro cultivation of bone marrow and synovial fluids derived from MSCs.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.
