Tetrad Dissection

Tetrad dissection is a genetic technique that separates and analyzes the four haploid products generated by a single meiotic event, allowing researchers to observe inheritance at the level of individual spores. In fungi such as yeast, a micromanipulator removes the spores from an ascus and places them separately on growth medium, where each develops into a genetically distinct colony. Comparing their traits reveals allele segregation, gene linkage, crossing over, and recombination patterns. In biology, tetrad dissection provides direct evidence of meiotic mechanisms and supports genetic mapping, mutant analysis, and studies of chromosome behavior.

The zebrafish kidney is home to both renal and hematopoietic adult stem/progenitor cells, and represents an outstanding opportunity to study these cell types and their progeny in a vertebrate model organism. Here, we demonstrate a detailed dissection procedure that enables the researcher to identify and surgically remove the adult zebrafish kidney, which can be used for applications such as cell isolation, transplantation, and expression studies of kidney and/or blood cell populations.

Micromanipulation of yeast cells is needed for meiotic genetic analysis or to select diploid zygotes. These micromanipulations are carried out using the microneedle of a dissection microscope. The microneedle is used to relocate cells and is controlled by a micromanipulator which are available with various degrees of automation.
