Tilt Series

A tilt series is a sequence of images acquired from the same specimen at different viewing angles, providing the projections needed to reconstruct its three-dimensional structure. In biological electron tomography, the specimen is incrementally tilted within an electron microscope while images are recorded, then aligned and computationally combined through tomographic reconstruction. This approach reveals the spatial organization of cellular compartments, membranes, macromolecular assemblies, and other structures that may be difficult to interpret in individual two-dimensional images. Tilt-series imaging supports ultrastructural analysis of cells and tissues, helping researchers connect molecular organization with biological function and disease-related changes.

The video demonstrates the generation of tilted amygdala slices from a mouse brain. The brain is positioned at an angle on the vibratome specimen stage to obtain slices containing the amygdala and axons from the medial prefrontal cortex that form synaptic connections with the amygdala. The brain slices are maintained at the physiological temperature to preserve the tissue for further experiments.
