Tissue Factor Role

Tissue factor is a transmembrane glycoprotein that initiates the blood-coagulation cascade and helps connect tissue injury with hemostasis. When exposed at damaged sites, it binds coagulation factor VII or VIIa, forming a tissue factor–factor VIIa complex that activates factors X and IX, leading to thrombin generation and fibrin formation. Its activity is normally separated from circulating blood by intact vascular barriers, limiting unwanted clotting. Understanding the tissue factor role is important in biology and medicine because dysregulated expression can contribute to thrombosis, inflammation, and tumor-associated coagulation, while also informing research on bleeding disorders and therapeutic anticoagulation.

Here we describe an in-house extracellular vesicle tissue factor activity assay. Activity-based assays and antigen-based assays have been used to measure tissue factor in extracellular vesicles from human plasma samples. Activity-based assays have higher sensitivity and specificity than antigen-based assays.
