Tissue Homogenization

Tissue homogenization is the process of mechanically or chemically disrupting biological tissue to produce a uniform sample, making its cells and molecular components accessible for analysis. Methods such as rotor-stator blending, grinding, bead beating, or sonication apply shear, impact, or ultrasonic energy, often in a chilled buffer that limits heat and preserves proteins, nucleic acids, and organelles. The resulting homogenate supports biochemical assays, DNA and RNA extraction, protein analysis, enzyme measurements, and organelle isolation. In biology, careful control of tissue type, buffer composition, and disruption intensity improves sample consistency and helps researchers connect molecular findings with tissue structure and function.

We describe a technique for the preparation of clarified human cortical homogenates, protein separation by SDS-PAGE, antigen retrieval and immunoblotting with an antibody to the Aβ peptide. Using this protocol, we consistently detect monomeric and multimeric Aβ in cortical tissue from humans with Alzheimer's pathology.
