Tissue Staining

Tissue staining is a set of methods that uses dyes or labeled probes to make specific structures in biological specimens visible under a microscope. It works through selective chemical interactions, such as dye binding to nucleic acids or proteins, or antibodies recognizing target molecules in immunohistochemistry. By revealing cell boundaries, nuclei, connective tissue, microorganisms, or molecular markers, staining helps researchers examine tissue organization and cellular activity. In biology, these approaches support histology, developmental studies, disease investigation, and the evaluation of experimental treatments, while standardized protocols improve comparison across samples.

Histological Staining of Neural Tissue

In order to examine the cellular, structural and molecular layout of tissues and organs, researchers use a method known as histological staining. In this technique, a tissue of interest is preserved using chemical fixatives and sectioned, or cut into very thin slices. A variety of staining techniques are then applied to provide contrast to the visually uniform sections. In the study of neuroanatomy, histological techniques are frequently applied to visualize and study nervous system tissue.

In this video, we describe a technique of hematoxylin and eosin staining of paraffin-embedded tissue sections to study their histology. The procedure includes deparaffinization of the tissue followed by staining the cells in the tissue. Upon staining, the nucleus appears distinctly blue, and the proteins in the cytoplasm display pink coloration.
