Tma Spot Positioning

TMA spot positioning is the organized placement of tissue cores within a tissue microarray (TMA) block, allowing many specimens to be analyzed on a single slide. A coordinate map guides the selection, spacing, and orientation of cores transferred from donor blocks into defined positions in a recipient block, linking each spot to its specimen and clinical or experimental metadata. In biology, accurate positioning supports consistent histological staining, immunohistochemistry, and molecular assays across large sample sets. It improves throughput, conserves valuable tissue, and enables standardized comparisons of biomarker expression, disease-associated changes, and treatment responses.

The purpose of the Specimen Orientation Tag (SpOT) is to function as an orientation tool to aid in individual tissue identification in multi-tissue paraffin blocks. These protocols demonstrate how it is constructed easily from common, low-cost histology materials and serves as a reliable visual marker in paraffin blocks and sections.

Finding Your Blind Spot and Perceptual Filling-in

Source: Laboratory of Jonathan Flombaum—Johns Hopkins University In the back of everyone's eye is a small piece of neural tissue called the retina. The retina has photosensitive cells that respond to stimulation by light. The responses of these cells are sent into the brain through the optic nerve, a bundle of neural fibers. In each retina there is a place somewhere in the periphery where the outputs from retinal cells collect and the bundled optic nerve exits to the brain. At that location,...

The preparing and processing of dried blood spots (DBS) for their final analysis are still poorly standardized for most diagnostic applications. To overcome this shortcoming, a comprehensive step-by-step protocol is suggested and subsequently evaluated with regard to its effectiveness for detecting markers of viral infections.

Light Spot Assay: A Method to Investigate Drosophila Phototactic Behavior

2023

Drosophila larvae are light-sensitive and respond to changes in surrounding light. This video describes an assay that tests their ability to avoid light, called the light spot assay. The featured protocol demonstrates how to set up the assay with a blue light LED and shows how to record the animal's behavior in response to the light encounter.
