Trna Mutations

tRNA mutations are changes in the genes or RNA sequences that produce transfer RNAs, molecules that deliver specific amino acids during protein synthesis. By altering tRNA structure, anticodon recognition, processing, stability, or interactions with aminoacyl-tRNA synthetases and ribosomes, these variants can disrupt accurate translation and reduce the production of functional proteins. Their effects help explain how genetic changes contribute to cellular dysfunction and inherited disease, while also revealing essential principles of gene expression. Studying tRNA mutations supports research in molecular genetics, disease mechanisms, and potential therapeutic strategies that restore translation or compensate for defective tRNA activity.

This protocol describes the preparation of a synthetic tRNA substrate for the Entamoeba histolytica DNA/tRNA methyltransferase 2 (Dnmt2) homolog Ehmeth and the measure of its methyltransferase activity. This experimental approach can be used for investigating the activity of other Dnmt2 proteins.
