Trypan Blue Assay

The Trypan blue assay is a dye-exclusion method used to estimate cell viability by distinguishing living cells from those with compromised membranes. When mixed with a cell suspension, Trypan blue enters cells whose plasma membranes have lost integrity, staining them blue, while viable cells exclude the dye and remain mostly unstained. Researchers count stained and unstained cells, often with a hemocytometer or automated counter, to calculate viability and cell concentration. In biology, the assay supports routine cell culture quality control, evaluation of cell proliferation or toxicity, and preparation of healthy populations for experiments such as transfection, drug testing, and tissue engineering.

Gut Acidification Monitoring Assay: A Technique to Study the Acidification of the Intestinal Lumen in Drosophila using Bromophenol Blue Dye

This video demonstrates an assay to monitor gut acidification in Drosophila flies. After the ingestion of food supplemented with a pH indicator dye, the acidification of the intestinal lumen results in a change in the color of the dye. Visual inspection of the digestive tract of the flies to examine the color change provides evidence of acidification.

Exfoliation of Egyptian Blue and Han Blue, Two Alkali Earth Copper Silicate-based Pigments

The preparation and exfoliation of CaCuSi4O10 and BaCuSi4O10 are described. Upon stirring in hot water, CaCuSi4O10 spontaneously exfoliates into monolayers, whereas BaCuSi4O10 requires ultrasonication in organic solvents. Near infrared (NIR) imaging illustrates the NIR emission properties of these materials, and aqueous dispersions of these nanomaterials are useful for solution processing.

This protocol describes the synthesis of biofunctionalized Prussian blue nanoparticles and their use as multimodal, molecular imaging agents. The nanoparticles have a core-shell design where gadolinium or manganese ions within the nanoparticle core generate MRI contrast. The biofunctional shell contains fluorophores for fluorescence imaging and targeting ligands for molecular targeting.
