Upright Mounting

Upright mounting is a specimen-preparation technique that positions a biological sample in a vertical or anatomically intended orientation for microscopy, helping preserve spatial relationships and improve image interpretation. The sample is stabilized on a slide, holder, or support using an appropriate mounting medium or restraint, while its orientation is maintained during preparation and observation. This approach is useful for examining tissues, embryos, organisms, and other structured specimens whose form or behavior depends on orientation. By reducing distortion and clarifying anatomical organization, upright mounting supports consistent imaging, comparative analysis, and documentation in biology and microscopy-based research.

We describe a protocol to surgically expose and stabilize the murine submandibular salivary gland for intravital imaging using upright intravital microscopy. This protocol is easily adaptable to other exocrine glands of the head and neck region of mice and other small rodents.
