Vascular System Development

Vascular system development is the biological process through which blood vessels form, expand, and organize into a functional network that supplies tissues with oxygen and nutrients while removing waste. During embryogenesis, endothelial precursor cells differentiate and assemble into primitive vascular tubes through vasculogenesis, while angiogenesis remodels and extends existing vessels through endothelial-cell sprouting, migration, and lumen formation. This tightly regulated process establishes circulation and tissue patterning and provides a foundation for studying congenital vascular disorders, tumor growth, wound healing, and regenerative medicine.

To date, thick tissue defects are typically reconstructed by applying autologous tissue flaps or engineered tissues. In this protocol, we present a new method for engineering vascularized tissue flap bearing an autologous pedicle, to serve as a substitute to autologous flaps.

Tools used for visualizing vascular regeneration require methods for contrasting the vascular trees. This film demonstrated a delicate injection technique used to achieve optimal contrasting of the vascular trees and illustrate the potential benefits resulting from a detailed analysis of the resulting specimen using µCT and histological serial sections.

Microfluidic Bioprinting for Engineering Vascularized Tissues and Organoids

We provide a generalized protocol based on a microfluidic bioprinting strategy for engineering a microfibrous vascular bed, where a secondary cell type could be further seeded into the interstitial space of this microfibrous structure to generate vascularized tissues and organoids.
