Vesicle Formation

Vesicle formation is the cellular process by which small, membrane-bound compartments bud from one membrane and transport materials to another, supporting organization and communication within cells. It begins when coat proteins and associated factors select cargo, concentrate it at a membrane site, and bend the lipid bilayer into a bud; membrane scission then releases the vesicle for movement and fusion with a target membrane. In biology, this process enables secretion, endocytosis, intracellular trafficking, and delivery of proteins and lipids to specific organelles. Understanding vesicle formation helps explain membrane homeostasis, cell signaling, development, and diseases linked to trafficking defects.

Microfluidic jetting against a droplet interface lipid bilayer provides a reliable way to generate vesicles with control over membrane asymmetry, incorporation of transmembrane proteins, and encapsulation of material. This technique can be applied to study a variety of biological systems where compartmentalized biomolecules are desired.

This video demonstrates the in vitro study of immunological synapse formation between an antigen-presenting cell and helper T lymphocytes using a fluorescence microscope. T cell receptors of helper T lymphocytes interact with an antigen-MHC-II complex in antigen-presenting cells and form immunological synapses, followed by secretory vesicle movements toward the synapse, which is visualized by fluorescent microscopy.
