Vesicle Preparation

Vesicle preparation is the process of producing membrane-bound compartments, such as liposomes or extracellular vesicle preparations, for studying biological membranes and their functions. In synthetic systems, amphipathic lipids self-assemble in aqueous buffer into bilayer spheres, while hydration, sonication, extrusion, or size-exclusion methods control vesicle formation, size, and composition. Prepared vesicles serve as models for membrane transport, signaling, fusion, and permeability, and can encapsulate proteins, nucleic acids, or small molecules for research and delivery studies. Careful control of lipid composition, buffer conditions, particle size, and purification improves reproducibility and enables comparisons across experiments.

This protocol describes an extrusion method for preparing lipid vesicles of sub-micron sizes with a high degree of homogeneity. This method uses a pressure-controlled system with controlled nitrogen flow rates for liposome preparation.
