Volume Acquisition

Volume acquisition is the process of capturing a series of images through the depth of a biological specimen to represent its three-dimensional structure. In fluorescence and other optical microscopy methods, the instrument records sequential optical sections as the focal plane or sample moves along the z-axis, producing a z-stack that can be reconstructed into a volumetric image. Appropriate spacing, exposure, and signal quality determine the resolution and completeness of the dataset. Volume acquisition supports quantitative analysis of cell organization, tissue architecture, developmental changes, and anatomical structures, enabling researchers to measure spatial relationships that cannot be assessed reliably from a single two-dimensional image.

We describe the preparation of ribbons of serial sections and their collection on large transfer support for use as Array Tomography samples, along with automated imaging procedures in a scanning electron microscope. The protocol allows screening, retrieval, and targeted imaging of local, rare events, and the acquisition of large data volumes.
