Wastewater Dna Extraction

Wastewater DNA extraction is the process of isolating genetic material from sewage or other wastewater samples so biological targets can be detected and studied. Because wastewater contains dilute DNA mixed with solids, organic matter, and substances that can inhibit analysis, the workflow typically concentrates material by filtration or centrifugation, breaks open cells through lysis, and purifies DNA for downstream testing. Researchers can then use methods such as polymerase chain reaction or sequencing to identify pathogens, monitor antimicrobial-resistance genes, and assess microbial communities. Reliable extraction supports wastewater-based epidemiology by turning complex environmental samples into measurable evidence of biological activity and public-health trends.

Enhanced Extraction of Low-Molecular Weight DNA from Wastewater for Comprehensive Assessment of Antimicrobial Resistance

Here, we present a simple technique to assess environmental antimicrobial resistance (AMR) by enhancing the proportion of low-molecular-weight extracellular DNA. Prior treatment with 20%-30% PEG and 1.2 M NaCl allows detection of both genomic and horizontally transferred AMR genes. The protocol lends itself to a kit-free process with additional optimization.

An experimental methodology is presented to compare the performance of small (100 L) and large (1,000 L) scale reactors designed for algae remediation of landfill wastewater. System characteristics, including surface area to volume ratio, retention time, biomass density, and wastewater feed concentrations, can be adjusted based on application.

Extraction of High Molecular Weight DNA from Microbial Mats

We provide an improved protocol for extracting high molecular weight DNA from hypersaline microbial mats. Microbial cells are separated from the mat matrix prior to DNA extraction and purification. This enhances the concentrations, quality, and size of the DNA. The protocol may be used for other refractory samples.

DNA extraction from saliva can provide a readily available source of high molecular weight DNA, with little to no degradation/fragmentation. This protocol provides optimized parameters for saliva collection/storage and DNA extraction to be of sufficient quality and quantity for downstream DNA assays with high quality requirements.
