Water Bath Incubation

Water bath incubation is a laboratory method that maintains samples at a controlled temperature by surrounding their containers with heated water, providing stable and uniform conditions for biological reactions. The bath transfers heat evenly through the vessel, while a thermostat regulates water temperature and minimizes fluctuations that can affect enzymes, cells, or other temperature-sensitive materials. In biology, researchers use water baths to incubate cultures, warm reagents, support enzyme assays, and maintain samples during procedures requiring gentle, consistent heating. This method helps improve experimental reproducibility and reduces the risk of overheating that can occur with direct heating sources.

Here, we present a protocol to demonstrate the generation of ice when water is introduced to a cold bath of brine, as a secondary refrigerant, at a range of temperatures well below the freezing point of water. It can be used as an alternative way of producing ice for industry.

The present protocol describes the maceration and cleaning of cadaveric bone with a vacuum-sealed, hot water bath immersion technique. This is a low-cost, safe, and effective method to produce anatomical specimens for surgical planning and medical education as an alternative to three-dimensional (3D) printed models.
