Z-rex Platform

The Z-REX Platform is a chemical biology method for delivering reactive electrophiles to selected proteins, enabling researchers to study how these small molecules alter protein function. It uses a genetically encoded HaloTag fusion to position a zinc-responsive electrophile precursor near a target protein; zinc activation releases the electrophile, which can covalently modify nearby nucleophilic residues, particularly cysteines. In biology, Z-REX helps map protein S-electrophilations, identify redox-sensitive signaling pathways, and connect electrophile exposure with changes in cellular activity. By providing spatially controlled chemical perturbation, the platform supports mechanistic studies of redox regulation, stress responses, and potential therapeutic targets.

Presented here is a method to mechanically phenotype single cells using an electronics-based microfluidic platform called mechano-node-pore sensing (mechano-NPS). This platform maintains moderate throughput of 1-10 cells/s while measuring both the elastic and viscous biophysical properties of cells.

A method for overcoming the optical diffraction limit is presented. The method includes a two-step process: optical phase retrieval using iterative Gerchberg-Saxton algorithm, and imaging system shifting followed by repetition of the first step. A synthetically increased lens aperture is generated along the direction of movement, yielding higher imaging resolution.

This technique can be applied to live fish of different developmental stages. Here, we couple standard assays in zebrafish with Z-REX for signaling pathway analysis.
