Human Esophageal Fibroblasts

Human esophageal fibroblasts are connective-tissue cells isolated from the esophagus that maintain the extracellular matrix and support tissue structure and repair. They produce and remodel matrix proteins, respond to mechanical and biochemical cues, and communicate with neighboring epithelial and immune cells through secreted factors and direct interactions. In cancer research, these cells provide a model for examining how the esophageal stromal microenvironment influences tumor initiation, invasion, and treatment response. Comparing normal fibroblasts with cancer-associated fibroblasts can reveal changes in cell signaling, matrix remodeling, and tumor-supportive behavior, helping identify mechanisms and potential therapeutic targets in esophageal cancer.

This manuscript describes the production, characterization and potential uses of a tissue engineered 3D esophageal construct prepared from normal primary human esophageal fibroblast and squamous epithelial cells seeded within a de-cellularized porcine scaffold. The results demonstrate the formation of a mature stratified epithelium similar to the normal human esophagus.

Carcinoma-associated fibroblasts (CAFs) rich in myofibroblasts present within the tumour stroma, play a major role in driving tumour progression. We developed a coimplantation tumour xengraft model for experimentally generating CAFs from human mammary fibroblasts. The protocol describes how to establish CAF myofibroblasts that acquire an ability to promote tumourigenesis.
