Cell Size Sorting

Cell size sorting is the separation of cells according to physical dimensions, a method that helps characterize cellular heterogeneity in cancer research. It works by passing a mixed cell population through a size-selective process, such as filtration, centrifugation, or microfluidic flow, in which cell diameter and deformability influence movement and collection. Researchers use the resulting fractions to compare tumor cells with surrounding or normal cells, enrich populations for downstream imaging and molecular analysis, and investigate how size variation relates to proliferation, differentiation, invasion, or treatment response. This approach can also support analysis of rare cell populations and improve the study of tumor evolution.

A method for isolating specific cell types from plant material is demonstrated. This technique employs transgenic marker lines expressing fluorescent proteins in particular cell types, cellular dissociation and Fluorescence Activated Cell Sorting. Additionally, a growth setup is established here that facilitates treatment of Arabidopsis thaliana seedlings prior to cell sorting.

In this video, we describe a protocol for the flow cytometry-based sorting of naïve B cell, memory B cell, and plasmablast plasma cell subpopulations from human peripheral blood B cells. The different B cell subpopulations can be distinguished based on fluorophore-based antibody binding to the CD19, CD27, and CD38 receptors expressed on the B cell surfaces, followed by cell sorting.
