Functional Resistance Assay

A functional resistance assay is a laboratory method that measures how effectively cancer cells withstand a treatment, providing a direct assessment of drug response beyond genetic predictions. In a typical assay, tumor cells or patient-derived samples are exposed to defined drug concentrations, and changes in cell viability, proliferation, or other functional readouts are quantified and compared with untreated controls. These assays can identify resistant or partially responsive cell populations, reveal treatment-specific vulnerabilities, and support the evaluation of candidate therapies. In cancer research, they help investigate resistance mechanisms and inform drug development and personalized treatment strategies.

2015

Drug resistance to targeted therapeutics is widespread and the need to identify mechanisms of resistance--prior to or following clinical onset--is critical for guiding alternative clinical management strategies. Here, we present a protocol to couple derivation of drug-resistant lines in vitro with sequencing to expedite discovery of these mechanisms.

In pressure myography, an intact small segment of a vessel is mounted onto two small cannulas and pressurized to a suitable luminal pressure. Here, we describe the method to measure vasorelaxation response of the mouse 3rd order mesenteric arteries in c57 and sGCα1-/- mice using pressure myography.

Seedling Flood Assay for Screening Bacterial Resistance in Tomatoes

Source: Hassan, J. A., et al. High-Throughput Identification of Resistance to Pseudomonas syringae pv. Tomato in Tomato using Seedling Flood Assay. J. Vis. Exp. (2020).This video demonstrates a method to screen for bacterial resistance in different tomato strains using ten-day-old seedlings inoculated with a bacterial suspension. It explains how bacteria invade through stomata, deliver effector proteins to suppress plant immunity, and how plant responses differ between susceptible and resistant...
