Fc Block Reagent

An Fc block reagent is a laboratory reagent that reduces nonspecific antibody binding by occupying Fc receptors on immune cells before antibody staining. Typically containing immunoglobulins or engineered Fc receptor-binding components, it saturates receptors on cells such as monocytes, macrophages, dendritic cells, and some tumor-associated immune populations, preventing antibody Fc regions from attaching independently of the target antigen. In cancer research, Fc block reagents improve the specificity and signal-to-noise ratio of flow cytometry, immunofluorescence, and tissue staining assays used to characterize tumor and immune-cell populations. Consistent blocking supports more reliable biomarker measurement, immune profiling, and evaluation of therapeutic responses.

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.
