Fumaroyl Chloride Reaction

The fumaroyl chloride reaction is a chemical derivatization method that modifies amine-containing metabolites for more effective analytical detection, making it valuable in cancer metabolomics. In this reaction, an amine nucleophile attacks an acyl carbon in fumaroyl chloride, displacing chloride and forming a stable fumaroylated derivative under controlled reaction conditions. The resulting products can improve chromatographic separation and mass-spectrometric response, supporting the measurement of amino acids and other primary or secondary amines in biological samples. Dynamic measurement of chloride ions is presented. Transition time of an Ag/AgCl electrode, during a chronopotentiometric technique, can give the concentration of chloride ions in electrolyte. This method does not require a stable conventional reference electrode.

The goal of this manuscript is to demonstrate a straightforward method for the preparation of ynones from acyl chloride and potassium alkynyltrifluoroborate salt starting materials. The one-pot reaction proceeds rapidly in the presence of boron trichloride without exclusion of air and moisture.
