Luminal Epithelial Cells

Luminal epithelial cells are polarized cells that line the inner surfaces, or lumina, of ducts and glandular structures, where they separate tissue compartments from fluid-filled spaces. Their apical surfaces face the lumen and support secretion, absorption, and barrier formation, while basolateral junctions and interactions with neighboring cells maintain tissue organization and regulate signaling. In cancer research, these cells are studied to understand epithelial homeostasis, tumor initiation, and progression, particularly when altered polarity, differentiation, or growth control enables abnormal proliferation and invasion. Their molecular markers and three-dimensional culture models also help classify tumors and evaluate disease mechanisms.

Transfer of Mammary Gland-forming Ability Between Mammary Basal Epithelial Cells and Mammary Luminal Cells via Extracellular Vesicles/Exosomes

This protocol describes methods for purifying, quantitating, and characterizing extracellular vesicles (EVs)/exosomes from non-adherent/mesenchymal mammary epithelial cells and for using them to transfer mammary gland-forming ability to luminal mammary epithelial cells. EVs/exosomes derived from stem-like mammary epithelial cells can transfer this cell property to cells that ingest the EVs/exosomes.

Whole-cell Patch-clamp Recordings of Isolated Primary Epithelial Cells from the Epididymis

We present a protocol that combines cell isolation and whole-cell patch-clamp recording to measure the electrical properties of the primary dissociated epithelial cells from the rat cauda epididymides. This protocol allows for investigation of the functional properties of primary epididymal epithelial cells to further elucidate the physiological role of the epididymis.

In this video, we demonstrate the culturing of isolated mouse prostate epithelial cells using a matrix gel-ring based culture technique. This approach can provide complementary information about the differentiation capacity of prostate epithelial cells to form 3D glandular structures in response to genetic or pharmacological manipulation.
