Mammosphere Medium

Mammosphere medium is a specialized, typically serum-free cell-culture formulation used to support the growth of mammary epithelial cells as three-dimensional mammospheres, making it valuable in cancer research. Under nonadherent culture conditions, selected cells survive and proliferate in suspension, while defined nutrients and mitogenic supplements support sphere formation and help assess self-renewal capacity. Researchers use mammosphere medium to study breast cancer stem-like cells, tumor initiation, differentiation, and responses to anticancer treatments. Mammosphere formation assays can therefore provide functional evidence of stem-like behavior and help evaluate mechanisms that influence tumor growth and therapeutic resistance.

2026

This protocol describes the method for setting up the mammosphere formation assay to study Apoptosis Inhibitor 5 (Api5)- induced cancer stemness in non-tumorigenic breast epithelial cells. Lysates collected from primary generation mammospheres are analysed for different stemness markers to assess the induction of stem cell-like properties.
