Metabolomics Sample Preparation

Metabolomics sample preparation is the set of procedures used to preserve, extract, and concentrate small-molecule metabolites from biological specimens before analytical measurement. In cancer research, samples are rapidly quenched to halt metabolic activity, then homogenized and treated with solvents that precipitate proteins and extract polar or nonpolar metabolites; centrifugation, filtration, and internal standards help produce clean, comparable extracts for mass spectrometry or nuclear magnetic resonance analysis. Careful control of temperature, solvent composition, and processing time reduces degradation and technical variation. Reliable preparation enables profiling of tumor and biofluid metabolomes, supports biomarker discovery, reveals altered pathways, and improves comparisons among disease states and treatment responses.

The metabolomic profile of Mycobacterium tuberculosis is determined after growth in broth cultures. Conditions can be varied to test the effects of nutritional supplements, oxidants, and anti-tuberculosis agents on the metabolic profile of this microorganism. Procedure for extract preparation is applicable for both 1D 1H and 2D 1H-13C NMR analyses.

This protocol aims to preserve cell metabolites for single-cell mass spectrometry (SCMS) by combining volatile salt solution washing, rapid liquid nitrogen quenching, freeze-drying, and −8 °C storage.
