P53 Transactivation

P53 transactivation is the process by which the p53 tumor suppressor activates specific genes to regulate cellular responses to stress, making it central to cancer research. Following DNA damage or other stress signals, p53 is stabilized and modified, binds response elements in target gene promoters, and recruits transcriptional coactivators and chromatin-modifying proteins to initiate gene expression. The resulting programs can promote cell-cycle arrest, DNA repair, senescence, or apoptosis, depending on cellular context. Studying p53 transactivation helps researchers assess how mutations disrupt tumor suppression, evaluate p53 activity in experimental models, and identify strategies for restoring or enhancing its function in cancer therapy.

Hs578T breast cancer cells harboring the p53 V157F mutation exhibit significantly higher Thioflavin T fluorescence compared to MCF7 cells, indicating enhanced protein aggregation. Multipoint fluorescence measurements improve detection accuracy and reliability in identifying β-sheet-rich aggregates, underscoring the importance of aggregation-prone p53 mutations in cancer research and the development of therapeutic strategies.
