Tumor Cell Quantification

Tumor cell quantification is the measurement of cancer cell number, density, or proportion within a sample, providing a basis for evaluating tumor burden and biological responses. Researchers quantify cells by separating or imaging specimens, identifying tumor-specific features such as morphology or molecular markers, and applying counting, segmentation, or flow cytometry methods under defined conditions. In cancer research, these measurements support tumor growth analysis, treatment-response studies, invasion and metastasis assays, and characterization of heterogeneous cell populations. Reliable quantification improves comparisons between experimental groups and helps connect cellular changes with disease progression, therapeutic effectiveness, and mechanisms of cancer biology.

This protocol demonstrates how to generate fluorescently marked, genetically defined clonal tumors in the Drosophila eye/antennal imaginal discs (EAD). Quantification of Tumor Cell Adhesion in Lymph Node Cryosections

Here, we describe a simple and inexpensive method that allows the quantification of adhesive tumor cells to lymph node (LN) cryosections. LN-adherent tumor cells are readily identified by light microscopy and confirmed by a fluorescence-based method, giving an adhesion index that reveals the tumor cell-binding affinity to LN parenchyma.

Here, we describe a simple and accessible strategy for visualizing, quantifying, and mapping immune cells in formalin-fixed paraffin-embedded tumor tissue sections. This methodology combines existing imaging and digital analysis techniques with the purpose of expanding the multiplexing capability and multiparameter analysis of imaging assays.

Bioluminescence imaging is a well-known tool for localizing tumors and metastases, but quantification of these images often requires complex calculations and particular instruments. We describe the easy-to-use luminoscore method, based on precise acquisition conditions, requiring no calculations, and enabling tumor burden and treatment response to be monitored in mouse models.
