Dab Substrate Incubation

DAB substrate incubation is a chromogenic detection step used to visualize target proteins in tissue sections and cell samples, particularly in immunohistochemistry for cancer research. During incubation, horseradish peroxidase linked to an antibody catalyzes the oxidation of 3,3′-diaminobenzidine (DAB) in the presence of hydrogen peroxide, forming an insoluble brown reaction product at the target site. Researchers use the resulting signal to identify tumor-associated biomarkers, assess protein distribution, and compare expression patterns across normal and malignant tissues. Careful control of incubation time and reagent conditions helps produce clear, interpretable staining for diagnostic research, biomarker validation, and studies of cancer biology.

By presenting a sophisticated environmental chamber, we provide an accurate temperature control method to imitate the magnitude and amplitude of in situ soil temperature and improve the experimental design of soil incubation studies.

This video demonstrates a method for prolonged incubation of neuronal tissue of the retina. Eyecups containing retina are obtained from a mouse eye; the inner limiting membrane (ILM) is enzymatically digested to expose the neuronal cells of the retina, and either the treated eyecup or the isolated retina is transferred to a specialized incubator for prolonged incubation at a low temperature.
