Chromatogram

Chromatogram is a graphical record of a chromatographic separation, showing how detector response changes as chemical components elute from a sample. In chemistry, compounds move through a stationary phase with a mobile phase at different rates because of differences in polarity, adsorption, partitioning, size, or charge, producing distinct signals at characteristic retention times. Peak positions can support compound identification, while peak areas or heights can estimate relative or absolute amounts when the method is calibrated. Chromatograms therefore help researchers assess sample composition, purity, reaction progress, and the performance of analytical methods such as gas chromatography and high-performance liquid chromatography.

Untargeted metabolomics provides a hypothesis generating snapshot of a metabolic profile. This protocol will demonstrate the extraction and analysis of metabolites from cells, serum, or tissue. A range of metabolites are surveyed using liquid-liquid phase extraction, microflow ultraperformance liquid chromatography/high-resolution mass spectrometry (UPLC-HRMS) coupled to differential analysis software.

A multidimensional gas chromatography method for the analysis of dissolved hydrogen sulfide in liquid crude oil samples is presented. A Deans switch is used to heart-cut light sulfur gases for separation on a secondary column and detection on a sulfur chemiluminescence detector.
