Fluorescent Labeled Oligonucleotide

A fluorescent labeled oligonucleotide is a short, synthetic nucleic acid strand linked to a fluorophore, allowing specific DNA or RNA sequences to be detected through light emission. When the oligonucleotide hybridizes with a complementary target, the attached dye produces or changes a fluorescent signal that can be measured by fluorescence spectroscopy, microscopy, or real-time amplification instruments. Researchers use these probes to identify nucleic acids, monitor hybridization, quantify molecular interactions, and analyze reaction kinetics in chemistry and biochemistry. Their sequence-specific recognition and optical readout support applications in genetic analysis, diagnostics, biosensing, and studies of molecular structure and function.

We describe a multiplex method for the detection of microorganisms within a sample using oligonucleotide-coupled fluorescent beads. Amplicon from all organisms within a sample is hybridized to a panel of probe-coupled beads. A Luminex or Bio-Plex instrument is used to query each bead for bead type and hybridization signal.
