Water Insoluble

Water-insoluble substances are compounds that dissolve only minimally in water under specified conditions, making their solubility an important property in chemistry, materials science, and laboratory practice. Their behavior reflects a balance between the energy required to separate particles in the substance and the interactions formed with water; nonpolar molecules often dissolve poorly because they cannot form favorable hydrogen-bonding or dipole interactions, while crystal lattice strength can further limit dissolution. Recognizing water-insoluble compounds supports solvent selection, liquid-liquid extraction, precipitation, filtration, and formulation design. Solubility changes with temperature, pH, and chemical modification, enabling controlled separation and delivery of chemical species.

Here, we present a protocol for the isolation of increasingly insoluble/aggregated alpha-synuclein (α-syn) from post-mortem human brain tissue. Through the utilization of buffers with increasing detergent strength and high-speed ultracentrifugation techniques, the variable properties of α-syn aggregation in diseased and non-diseased tissue can be examined.

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.
