Drug Dosing Estimation

Drug dosing estimation is the process of determining an appropriate medication dose and schedule to achieve therapeutic effects while limiting toxicity. In clinical practice, it combines pharmacokinetic principles, such as absorption, distribution, metabolism, and excretion, with pharmacodynamic relationships between drug concentration and response; calculations may also account for body weight, age, renal or hepatic function, and therapeutic range. Accurate estimation supports individualized treatment, particularly for drugs with narrow therapeutic windows or substantial patient-to-patient variability. It guides initial prescribing, dose adjustment, and therapeutic drug monitoring, improving medication safety and helping clinicians balance efficacy with adverse effects.

The cytogenetic dicentric chromosome (DC) assay quantifies exposure to ionizing radiation. The Automated Dicentric Chromosome Identifier and Dose Estimator software accurately and rapidly estimates biological dose from DCs in metaphase cells. It distinguishes monocentric chromosomes and other objects from DCs, and estimates biological radiation dose from the frequency of DCs.

Drug Dosing in Renal Diseases: Estimation of Glomerular Filtration Rate Based on Serum Creatinine Concentration

2025

Glomerular filtration rate (GFR) can be estimated from serum creatinine using the modification of diet in renal disease (MDRD) formula or the chronic kidney disease–epidemiology collaboration (CKD–EPI) equation. Both methods are widely used in clinical practice to assess kidney function and guide treatment decisions. The MDRD equation does not require weight or height measurements and is normalized to the body surface area of 1.73 m², considered the average adult surface area. This equation is...

Dose Escalation: A Method for Developing Drug Resistance in Cancer Cells

2023

The video describes the development of drug resistance in cancer cells by exposing cells to escalating doses of a drug after calculating IC50 values of the drug previously with MTT assay. The sample protocol describes the step-by-step process of developing drug-resistant cells.

Drug Dosing: Infants and Children

Pediatric patient dosages diverge from adults due to disparities in body surface area, total body water, and extracellular fluid per kilogram of body weight. The dosing regimen considers the variations in pharmacokinetics and pharmacology across distinct age groups, encompassing preterm newborns, infants, young children, older children, and adolescents. Calculation of pediatric patient doses is predicated on determining body surface area, which exhibits a superior correlation with the child's...

Drug Dosing: Geriatric Patients

2025

Elderly individuals encompass a diverse population with varying degrees of age-related physiological changes. Defining the elderly presents challenges, as the geriatric population is often arbitrarily categorized as individuals older than 65. However, many individuals in this group lead active and healthy lives, with an increasing number surpassing 85 years and falling into the older elderly category. Physiological changes associated with aging impact performance capacity and homeostatic...
