Pancreatic Duct Obstruction

Pancreatic duct obstruction is a blockage that prevents pancreatic secretions from reaching the duodenum, disrupting digestion and potentially injuring the pancreas. Obstruction may result from gallstones, ductal stones, tumors, scar-related strictures, or congenital abnormalities; retained secretions increase ductal pressure and can trigger inflammation, including acute or chronic pancreatitis. Clinicians evaluate suspected obstruction using symptoms, laboratory tests, and imaging such as ultrasound, computed tomography, magnetic resonance cholangiopancreatography, or endoscopic retrograde cholangiopancreatography. Identifying the underlying cause guides treatment, which may include endoscopic stone removal or stenting, surgery, and management of complications such as infection, pain, or impaired digestion.

Disruption of bile flow results in severe inflammatory cholestatic liver injury with a characteristic time-dependent sequence of morphological alterations. Here we present a protocol for the surgical ligation of the common bile duct in mice that allows to induce a strong fibrotic response after 21 to 28 days.
