Gallstone Blockage

Gallstone blockage is a condition in which a gallstone obstructs the cystic duct or common bile duct, preventing normal movement of bile from the gallbladder into the intestine. Stones typically form when bile components crystallize in the gallbladder and may migrate into a duct, where they can trigger pressure, inflammation, jaundice, and abdominal pain; obstruction near the pancreatic duct may also cause pancreatitis. Clinically, recognizing this process guides evaluation with laboratory tests and imaging and supports treatments such as endoscopic stone removal or gallbladder surgery, helping prevent infection, tissue injury, and recurrent biliary disease.

Endolymphatic duct blockage is a relatively new surgical method for patients suffering from Ménière's disease. Following a regular mastoidectomy, the endolymphatic duct is identified and ligated using a regular titanium hemoclip. The effectiveness of this procedure is currently being assessed in a randomized trial.

We describe here a system utilizing a site-specific, reversible in vivo protein block to stall and collapse replication forks in Escherichia coli. The establishment of the replication block is evaluated by fluorescence microscopy and neutral-neutral 2-dimensional agarose gel electrophoresis is used to visualize replication intermediates.
