Cardiac Organoid Differentiation

Cardiac organoid differentiation is the process of guiding stem cells to self-organize into three-dimensional, heart-like tissues that model early cardiac development. In controlled culture, timed modulation of signaling pathways, particularly sequential activation and inhibition of Wnt signaling, directs pluripotent stem cells through mesoderm and cardiac progenitor stages, while cell-cell interactions promote tissue organization. These organoids can contain cardiomyocytes and other cardiac cell types, enabling researchers to study heart formation, lineage specification, and tissue maturation. In developmental biology, they support investigations of congenital heart disease, provide platforms for evaluating drug effects and cardiotoxicity, and offer a scalable model for testing regenerative strategies.

2025

In this video, a stepwise protocol for Human embryonic stem cells into neural organoids is demonstrated. The method involves inducing neuronal rosette formation, followed by controlled maturation and final culturing on a hydrophobic membrane to achieve fully developed neural organoids.

A fine tuning regulation of gene transcription underlies embryonic cell fate decision. Herein, we describe chromatin immunoprecipitation assays used to investigate epigenetic regulation of both cardiac differentiation of stem cells and cardiac development of mouse embryos.

In Vitro Resident Memory CD8 T Cell Differentiation Using Epithelial Organoid-T Cell Co-culture System

To study CD8 T cell differentiation and function in vitro, CD8 T cells can be isolated from the mouse to be co-cultured long-term with preformed infected murine vaginal epithelial organoids. Here, we describe this process and assess the acquisition of resident memory T cell markers upon co-culture.

Conventional methods to initiate suspension aggregate based cardiac differentiation of human pluripotent stems cells (hPSCs) are plagued with culture heterogeneity with respect to aggregate size and shape. Here, we describe a robust method for cardiac differentiation employing microwells to generate size-controlled hPSC aggregates cultured under cardiac-promoting conditions.
