Cell Type Composition

Cell type composition is the distribution and relative abundance of distinct cell types within a tissue, organ, or biological sample, a property that shapes its structure and function. During development, coordinated cell proliferation, differentiation, migration, and programmed cell death alter this composition, while signaling between neighboring cells helps establish and maintain cell identities. Measuring cell type composition with microscopy, flow cytometry, or single-cell sequencing allows researchers to track lineage formation, compare normal and perturbed development, and interpret tissue organization. These analyses also support studies of organoids, congenital disorders, regeneration, and how changes in cellular proportions influence developmental outcomes.

Immunohistochemical staining of myosin heavy chain isoforms has emerged as the state-of-the-art discriminator of skeletal muscle fiber-type (i.e., type I, type IIA, type IIX, type IIB). Here, we present a staining protocol along with a novel semi-automated algorithm that facilitates rapid assessment of fiber-type and fiber morphology.
