Drosophila Leg Development

Drosophila leg development is the process by which a fruit fly’s embryonic and larval cells form a segmented adult appendage, providing a powerful model for studying pattern formation and organogenesis. During larval growth, leg imaginal disc cells receive positional signals, including Wingless, Decapentaplegic, and Hedgehog, that establish body axes and specify structures such as the coxa, femur, tibia, and tarsus; hormonal cues then trigger disc eversion and metamorphic remodeling. Because these pathways are conserved across animals, Drosophila leg development helps researchers analyze gene regulation, cell signaling, tissue growth, and congenital patterning defects in developmental biology.

One of the many reasons that make Drosophila an extremely valuable organism is that the molecular, cellular, and genetic foundations of development are highly conserved between flies and higher eukaryotes such as humans. Drosophila progress through several developmental stages in a process known as the life cycle and each stage provides a unique platform for developmental research. This video introduces each stage of the Drosophila life cycle and details the physical characteristics and major...

Source: Guan, W., et. al. Visualize Drosophila Leg Motor Neuron Axons Through the Adult Cuticle. J. Vis. Exp. (2018) This video demonstrates imaging of Drosophila legs with green fluorescent protein-expressing motor neurons using confocal microscopy. A laser and two detectors capture GFP and cuticle autofluorescence. Images are processed by splitting channels, subtracting autofluorescence, and merging into a colored composite image to visualize axons within the leg segment.
